Hydrogen sulfide (H 2 S) plays crucial roles in antioxidation, anti-inflammation, and cytoprotection. Despite substantial progress the design synthesis of activatable H S donors, methods for high-precision detection imaging released living systems have been lacking. In this study, a biothiol-activated near-infrared (NIR) fluorescent donor, PRO-ST , was developed real-time visualization release. consists dicyanoisophorone-based NIR fluorescence moiety (TCOO), biothiol-trigger (4-isothiocyanate benzyl alcohol), sulfur-source group (thiophosgene). exhibits high enhancement (45-fold), outstanding release efficiency (73%), controllable (60 min), excellent cell compatibility. These distinctive features enable to be applied visualizing cells, zebrafish, mice. Moreover, performance anti-inflammation wound healing biological systems, as confirmed by situ Thus, provides versatile effective method detect visualize release, elucidate mechanisms underlying healing, or optimize interventional therapy.
